A PD-1 agonist, deliberately the opposite of what PD-1 antibodies do in cancer, that missed in alopecia areata and was dropped for it in 2023.

How it is meant to work
PD-1 is the brake on an activated T cell. Oncology antibodies release that brake so the immune system attacks a tumour; rosnilimab presses it, to quiet the T cells attacking the follicle. Monthly subcutaneous dosing was supported by the healthy-volunteer phase 1.
Why it matters
The mechanism had a striking piece of corroborating evidence behind it: PD-1 blockade in cancer patients is a recognised cause of alopecia areata, so a drug doing the reverse was a hypothesis whose human experiment had already been run in the other direction. The direct experiment then failed: in January 2023 AnaptysBio reported that among 51 patients no one on rosnilimab reached a SALT score below 20 at week 24, that absolute SALT scores did not support the target profile, and that development in alopecia areata would not be pursued. The registry entry was never updated.
