How it is meant to work
Daxdilimab targets ILT7 and depletes plasmacytoid dendritic cells, the cells responsible for type I interferon production. Those cells appear in the infiltrate around affected follicles, which is the argument for removing them rather than inhibiting the signalling downstream of them.
Why it matters
This is the most upstream intervention in the register for alopecia areata. A JAK inhibitor blocks a signal continuously and has to keep being taken; depleting the cell that produces the signal is a different bet about how the disease sustains itself. The phase 2 has completed and no efficacy data has been published, which is itself worth noting.
