How it is meant to work
Aldena builds siRNA for skin. The trial record does not name the gene ALD-102 silences; Alys Pharmaceuticals, Aldena's parent, announced the same trial in March 2025 under the code ALY-101 and said it targets JAK1, with preclinical work at UMass Chan. The idea is the effect of an oral JAK inhibitor, confined to where the needle goes.
Why it matters
Oral JAK inhibitors, the approved option for severe alopecia areata in the US, EU and Japan, act on the whole body; a gene-silencing injection that acts only where it is placed would sidestep that, if it works and if patients accept intradermal injections. ARISE, the Phase 1b/2a, has recruited in the US and Canada since March 2025 and treats areas of the same scalp with drug, placebo or nothing; primary completion is expected in October 2026. Aldena is one of the six companies Medicxi combined into Alys Pharmaceuticals in 2024.
