NEOSH101

Development of NEOSH101 for androgenetic alopecia was discontinued by Neosil.

A topical proteasome inhibitor that reported a 4.8% hair-count gain in 2007 and was shelved by its acquirer two years later.

How it is meant to work

Registered as a tetrapeptide aldehyde proteasome inhibitor applied to the scalp. The registry gives no account of how proteasome inhibition was expected to act on the follicle.

Why it matters

A Phase 2a in Germany, 50 men, two 14-day courses of once-daily NEOSH101 or placebo a month apart, reported a 4.8% increase in total hair count and 3.7% in cumulative thickness in a January 2007 company release that did not say what the gains were measured against, and that never became a paper this register can find. Two larger studies registered in 2007, one of 180 men against 5% minoxidil and placebo that completed in April 2008 and a dose-ranging study of 140, posted no result. Peplin bought Neosil in October 2008 and wrote in its 2009 annual report that it was not conducting further development of the Neosil intellectual property; LEO Pharma agreed to buy Peplin that September, and the compound has not surfaced since. One of the earlier novel-mechanism topicals to disappear inside an acquisition.
